چکیده
Adeno-associated virus (AAV) is one of the most promising gene transfer vector types featuring long-term expression and low toxicity. The lack pathogenicity availability many serotypes augmented applicability AAV virions in therapy applications. recombinant capsid includes therapeutic protein-coding transgene as well a promoter to initiate translation poly A sequence portion for stabilization. Current manufacturing technologies, however, cannot guarantee generation only full capsids, i.e., including entire required genome. Partially filled empty capsids are also part product, decreasing this way efficacy safety upon clinical translation. Therefore, rapid, accurate QC friendly analysis ratio high importance during release testing. In paper, an automated capillary isoelectric focusing technique introduced, readily applicable biopharmaceutical industry fast efficient determination ratio. method reveals information about proportion partially capsids. For higher resolution (<0.1 pI unit), mixtures wide narrow range ampholytes were utilized. point peak area percentage reproducibility (RSD) mixed ampholyte assay 1.67% 2.45 %, respectively, requiring 65 nL sample volume per injection.

متن کاملQuantification of hemoglobin variants by capillary isoelectric focusing.
Capillary isoelectric focusing (cIEF) was used to identify and quantify major and minor hemoglobin (Hb) variants. Whole blood (approximately 10 microL required) hemolysate was analyzed with a commercial instrument equipped with a 50 microns (i.d.) x 27 cm coated capillary filled with 20 g/L ampholytes (pH 6-8) in 4 g/L methylcellulose (MC). Cathode and anode solutions were 20 mol/L NaOH and 100...
متن کاملProteolytic mapping of the adeno-associated virus capsid.
The three-dimensional structures of the viral capsid of three AAV serotypes have previously been determined by X-ray crystallography or cryoelectron microscopy. These studies of AAV and similar studies of autonomous parvoviruses have yielded important structural information about the virions in a low-energy conformation. However, there is little information on the structural properties of AAV v...
متن کاملGeneration of Helper Plasmids Encoding Mutant Adeno-associated Virus Type 2 Capsid Proteins with Increased Resistance against Proteasomal Degradation
Objective(s): Adeno-associated virus type 2 (AAV2) vectors are widely used for both experimental and clinical gene therapy. A recent research has shown that the performance of these vectors can be greatly improved by substitution of specific surface-exposed tyrosine residues with phenylalanines. In this study, a fast and simple method is presented to generate AAV2 vector helper plasmids encod...
متن کاملAdeno-associated virus proteins: origin of the capsid components.
The three primary capsid proteins (A, B, and C) of adeno-associated viruses have been shown previously to contain overlapping amino acid sequences (R. McPherson and J. Rose, J. Virol. 46:523-529, 1983). In the present study we demonstrate definitively that these proteins are encoded in the right half of the adeno-associated virus 2 genome, and one or both of the smallest adeno-associated RNA sp...
